摘要 目的:建立高相高效液相色谱法同时测定白芷香豆素提取物中花椒毒酚、8-甲氧基-5-羟基补骨脂素、水合氧化前胡素、佛手柑内酯、欧前胡素、蛇床夫内酯、异欧前胡素7种成分在大鼠血浆内的含量。方法:色谱柱为Agilent TC-C 18 (250 mm×4.6 mm,5 μm);流动相:以甲醇为流动相A,纯水为流动相B,梯度洗脱;流速: 1.0 ml/min;检测波长:310 nm。结果:花椒毒酚、8-甲氧基-5-羟基补骨脂素、佛手柑内酯在0.032~20 μg/ml范围内线性关系良好,水合氧化前胡素、欧前胡素、蛇床夫内酯、异欧前胡素在 0.016~20 μg/ml范围内线性关系良好,精密度、准确度良好,回收率符合标准。结论:该方法同时测定了白芷香豆素提取物中7种成分在大鼠血浆内的含量,方法快速、简便,具有较好的精密度和准确度,为白芷体内药代学研究提供数据学参考。 Objective: A high-performance liquid chromatography (HPLC) method was established for simultaneous determination of seven components (including xanthotoxol, 5-hydroxy-8-methoxypsoralen, oxypeucedanin hydrate, bergapten, imperatorin, cnitidin, and isoimperatorin) in Angelica coumarins extract in rat plasma. Methods: The separation was performed on an Agilent TC-C 18 (250 mm×4.6 mm, 5 μm) with the gradient elution of methanol(A) and water (B)(0~30 min,35%-90% A;30.5~40 min,35% A) at flow rate of 1.0 ml/min. Detection wavelength was set at 310 nm.Results: Within the range of 0.032~20 μg/ml, the concentrations of xanthotoxol, 5-hydroxy-8-methoxy psoralen and bergapten showed a great linear relationship. The linear range of oxypeucedanin hydrate, imperatorin, cnitidin and isoimperatorin was 0.016~20 μg/ml, respectively. The established HPLC method showed good precision and accuracy, and the recovery rate met the standards. Conclusion: In this paper, we have established a simple and accurate HPLC method for simultaneous determination of seven components in the extract of Angelica coumarins in rat plasma. The method is rapid, simple, and has good precision and accuracy, which can provide data reference for the pharmacokinetic study of Angelica dahurica in vivo.
