摘要
SOX基因家族是一系列在性别分化、胚胎发育、细胞多功能性的维持等方面具有重要作用的转录因子,在高等脊椎动物中研究比较深入,但在淡水珍珠蚌中却很少见。本研究利用RACE法克隆获得了三角帆蚌SOX2基因的序列,其cDNA全长为1840bp,开放阅读框为672bp,编码氨基酸223个。该基因具有SOX基因家族典型的HMG-box蛋白结构域,与其他物种对比发现,保守性极高。实时荧光定量PCR组织表达结果显示,在斧足中表达量最高,在肝脏中最低,同时发现雌雄性腺之间存在极显著性差异,雌性性腺表达量明显高于雄性性腺;性腺在早期胚胎中表达量明显高于其他时期,推测SOX2基因参与了三角帆蚌的性别分化和胚胎发育过程。
SOX family is a series of transcription factors that play an important role in sex differentiation, embryonic development, and maintenance of cells pluripotency. There are numerous reports on vertebrates, but rare in freshwater bivalve. In this study, the sequence of SOX2 gene of Hyriopsis cumingii was cloned by RACE. The full length of cDNA was 1 840 bp. The open reading frame was 672 bp, encoding 223 amino acids. The gene has a typical HMG-box protein domain of SOX gene family and is highly conserved compared with other species. Quantitative Real-time RCR detected that the highest expression was in foot and the lowest in liver. Meanwhile, there is a significant difference in gonad between male and female. The expression of female gonad was significantly higher than that of male gonad. The expression of early gonad periods was found that early embryo was significantly higher than other periods. It is speculated that the SOX2 gene is involved in the sex differentiation and embryonic development of H. cumingii.
