摘要
目的观察未破裂颅内动脉瘤(ICA)患者外周血中microRNA-126表达情况,探讨microRNA-126潜在靶基因及其在ICA发生中的可能机制。方法收集住院或体检的ICA患者120例作为ICA组,另选取体检无ICA且性别、年龄、伴随疾病与ICA组匹配者63例作为对照组。采用RT-PCR定量检测2组受检者外周血中microRNA-126的相对表达量,并采用UCSC数据库对microRNA-126的基因位点及同源性进行分析,采用TargetScan数据库及CoMeTa数据库对microRNA-126的潜在靶基因进行预测,采用DAVID数据库对microRNA-126的潜在靶基因的功能富集(GO富集)和KEGG通路富集进行分析。结果ICA组患者外周血中microRNA-126的相对表达量明显高于对照组(P<0.05);通过靶基因预测软件共获得包括ITGA6、CRK、PCDH7、ADAM9等在内的15个潜在靶基因;GO分析和KEGG通路分析显示microRNA-126靶基因功能主要富集于蛋白质结合的形成以及FAS信号通路中。结论microRNA-126在ICA患者中表达上调,并通过调控FAS信号通路中的多个靶基因参与ICA的发生。
Objective It is to observe the expression of microRNA-126 in peripheral blood of patients with unruptured intracranial aneurysms(ICA), and to explore the potential target genes of microRNA-126 and its possible mechanisms in the development of ICA. Methods 120 patients with ICA who were hospitalized or examined were enrolled in the ICA group. 63 patients without ICA in the body examination and their gender, age, concomitant disease matched with ICA group were selected as the control group. The relative expression of microRNA-126 in peripheral blood of the subjects in the two groups was detected by RT-PCR quantitative detection. The UCSC database was used to analyze the microRNA-126 gene locus and homology. The TargetScan database and CoMeTa database were used to predict the potential target genes of microRNA-126, and the DAVID database was used to analyze the functional enrichment(GO enrichment) and KEGG pathway enrichment of the potential target genes of microRNA-126. Results The relative expression of microRNA-126 in peripheral blood of ICA group was significantly higher than that of the control group(P<0.05). 15 potential target genes including ITGA6, CRK, PCDH7 and ADAM9 were obtained by target gene prediction software;GO analysis and KEGG pathway analysis revealed that the microRNA-126 target gene function was mainly enriched in the formation of protein binding and in the FAS signaling pathway. Conclusion MicroRNA-126 is up-regulated in ICA patients and participates in the development of ICA by regulating multiple target genes in the FAS signaling pathway.
