摘要
目的分析重度子痫前期患者血清和胎盘血管内皮生长因子(VEGF)、可溶性血管内皮生长因子受体1(s VEGFR-1)水平异常与长链非编码RNA-LOC391533(lnc RNA-LOC391533)表达的相关性。方法采用前瞻性研究,选取青海红十字医院2017年1月至2019年1月收治的80例重度子痫前期患者(病例组),同期选取45例产检正常的健康孕妇(对照组)。测量两组孕妇胎盘组织中螺旋动脉管腔面积和管壁厚度情况,通过酶联免疫吸附法测定两组孕妇血清中VEGF和s VEGFR-1含量,根据免疫印迹法测定孕妇胎盘组织中VEGF和s VEGFR-1蛋白的表达水平,采用实时荧光定量聚合酶链反应测定孕妇胎盘组织中lnc RNA-LOC391533 m RNA相对表达量。采用Pearson相关性分析病例组患者胎盘组织中lnc RNA-LOC391533 m RNA相对表达量与胎盘螺旋动脉管腔面积、管壁厚度、血清和胎盘组织中VEGF、s VEGFR-1水平的关系。结果相比对照组,病例组胎盘组织螺旋动脉管壁厚度明显增厚,管腔面积明显减少(P<0.01)。病例组血清和胎盘组织中VEGF的水平较对照组均明显下降,s VEGFR-1的水平较对照组均明显升高(P<0.01)。病例组胎盘组织中lnc RNA-LOC391533 m RNA相对表达量明显升高(P<0.01)。经Pearson相关性分析发现,病例组患者胎盘组织中lnc RNA-LOC391533 m RNA相对表达量与血清和胎盘组织中VEGF的水平、螺旋动脉管腔面积均存在负相关关系(P<0.01),与血清和胎盘组织中s VEGFR-1的水平、螺旋动脉管壁厚度均存在正相关关系(P<0.01)。结论重度子痫前期患者血清和胎盘组织中VEGF与s VEGFR-1蛋白表达异常,其原因可能与胎盘螺旋动脉重铸不足的发生存在密切关系。
Objective To analyze the correlation between abnormal serum and placental levels of vascular endothelial growth factor(VEGF)and soluble vascular endothelial growth factor receptor 1(sVEGFR-1)and the expression of long non-coding RNA-LOC391533(lncRNA-LOC391533)in patients with severe preeclampsia.Methods A prospective study was conducted to select 80 patients with severe preeclampsia(case group)admitted to our hospital from January 2017 to January 2019,and 45 healthy pregnant women(control group)with normal obstetric examination at the same time.The area and wall thickness of spiral artery in placenta were measured.The serum levels of vascular endothelial growth factor and sVEGFR-1 were measured by enzyme-linked immunosorbent assay.The expression levels of vascular endothelial growth factor and sVEGFR-1 protein in placenta of pregnant women were measured by immunoblotting.The pregnancy was measured by real-time fluorescence quantitative polymerase chain reaction.Relative expression of lncRNA-LOC391533 in placenta.Pearson correlation was used to analyze the relationship between the relative expression of lncRNA-LOC391533 in placenta tissue and the area of the lumen of the spiral artery,the thickness of the wall,the levels of vascular endothelial growth factor and sVEGFR-1 in serum and placenta.Results Compared with the control group,the spiral artery wall thickness of placenta tissue in the case group was significantly thicker and the lumen area was significantly reduced(P<0.01).The levels of vascular endothelial growth factor in serum and placenta of the case group were significantly lower than those of the control group,and the levels of sVEGFR-1 were significantly higher than those of the control group(P<0.01).The relative expression of lncRNA-LOC391533 in placenta tissue of case group was significantly increased(P<0.01).Pearson correlation analysis showed that the relative expression of lncRNA-LOC391533 was negatively correlated with the levels of vascular endothelial growth factor in serum and placenta tissues and the area of spiral artery lumen(P<0.01),and positively correlated with the levels of sVEGFR-1 in serum and placenta tissues and the thickness of spiral artery wall.Department(P<0.01).Conclusion The abnormal expression of VEGF and sVEGFR-1 in serum and placenta of patients with severe preeclampsia may be closely related to the occurrence of inadequate placental spiral artery recasting.
