miR-149-3p对糖尿病大鼠心肌细胞胰岛素抵抗的作用 被引量:1

摘要 目的探讨miR-149-3p对糖尿病大鼠心肌细胞胰岛素抵抗的干预作用及其机制。方法应用高脂饲料联合链脲佐菌素方法构建糖尿病大鼠模型,提取心肌组织总RNA,检测miR-149-3p的表达情况。培养H9c2心肌细胞,体外应用高糖高脂诱导H9c2细胞胰岛素抵抗模型,检测miR-149-3p的表达情况。应用miRNA转染技术将miR-149-3p转染进H9c2心肌细胞,并检测葡萄糖摄取。应用生物信息学实验、real time qPCR实验和western blot实验确定fat mass and obesity associated(FTO)是否为miR-149-3p的靶点。结果糖尿病模型组大鼠腹腔注射糖耐量实验中2h血糖显著高于对照组,且其血浆TC、TG和FFA水平均显著高于对照组。此外,糖尿病模型组大鼠心肌中miR-149-3p的表达显著降低;过表达miR-149-3p能够抑制胰岛素刺激的葡萄糖摄取,敲减miR-149-3p以能够促进胰岛素刺激的葡萄糖摄取。生物信息学实验、real time qPCR实验和western blot实验证明FTO是miR-149-3p的直接靶点。结论 miR-149-3p通过下调FTO抑制糖尿病大鼠心肌细胞胰岛素抵抗。
