间质性肺病小鼠循环纤维细胞与血清KL-6的相关性 被引量:2

摘要 目的观察博来霉素(BLM)诱导的间质性肺病(ILD)小鼠外周血循环纤维细胞的数量和血清Ⅱ型肺泡表面抗原(KL)-6水平。方法40只SPF级昆明小鼠随机分为2组:模型组(A组,n=20)和对照组(B组,n=20)。A组小鼠一次性气管滴注BLM5mg/kg;B组气管滴注等体积的生理盐水。分别于造模后第7、14、21、28天,流式细胞仪检测外周血中循环纤维细胞百分比,酶联免疫吸附试验检测血清KL-6浓度,苏木素-伊红(HE)染色和Masson染色观察肺组织炎症及肺纤维化程度,碱水解法测定羟脯氨酸含量。结果A组在BLM处理后第14天炎性症状最明显,第21天纤维化最明显,A组在第7、14、21天炎症评分显著高于B组(P<0.01),A组各个时期的纤维化评分均显著高于B组(P<0.01);A组各个时期羟脯氨酸含量均显著高于B组(P<0.01);A组外周血循环纤维细胞百分比在给药后第7天水平达到最高,之后呈下降趋势,A组在第7、14、21天均显著高于B组(P<0.01);A组血清KL-6浓度在第14、21、28天时均显著高于B组(P<0.05),第21天达到最高;A组和B组外周血循环纤维细胞数量与肺部炎症、纤维化评分、羟脯氨酸含量和血清KL-6浓度呈正相关(r=0.631、0.676、0.764、0.454,P<0.01)。结论在BLM诱导的小鼠ILD的病理进程中,外周血循环纤维细胞数量消长与肺部炎症和纤维化程度呈成正相关,循环纤维细胞可能参与ILD的发生发展,提示循环纤维细胞可作为ILD诊断的生物学标志物。
