长链非编码RNA linc00261在卵巢癌组织中的表达及其临床意义 被引量:5

摘要 目的探讨长链非编码RNA(LncRNA)linc00261在卵巢癌中的表达及其在卵巢癌发生发展中的可能作用机制。方法应用qRT-PCR检测60例原发性卵巢癌患者的卵巢癌组织及癌旁组织中LncRNA linc00261的表达,并分析其表达与卵巢癌患者临床病理特征的相关性。通过设计合成LncRNA linc00261表达质粒和对照质粒,转染SKOV3细胞,qRT-PCR检测转染细胞中LncRNA linc00261的表达水平,MTT法检测细胞增殖活性,划痕实验检测细胞迁移能力,Transwell小室法检测细胞侵袭能力。结果 qRT-PCR检测结果显示,卵巢癌组织中LncRNA linc00261的平均相对表达量(2.47±0.93)明显低于癌旁组织(6.82±1.56),差异具有统计学意义(t=5.153,P<0.05)。卵巢癌组织中LncRNA linc00261的表达与患者的年龄、是否绝经、病理类型无关(均P>0.05),但与患者的肿瘤组织分化程度、临床分期、有无淋巴结转移、有无腹水等有关(均P<0.05)。转染SKOV3细胞后,LncRNA linc00261表达质粒组的LncRNA linc00261表达水平显著高于对照质粒组和空载体组(均P<0.05);MTT检测结果显示,转染48、72、96 h后,LncRNA linc00261表达质粒组SKOV3细胞增殖能力显著低于对照质粒组和空载体组(均P<0.05);划痕实验显示,LncRNA linc00261表达质粒组细胞迁移能力显著低于对照质粒组和空载体组(均P<0.05);侵袭实验的结果显示,LncRNA linc00261表达质粒组穿膜细胞数显著低于对照质粒组和空载体组(均P<0.05)。结论 LncRNA linc00261在卵巢癌患者癌组织中表达下调,其表达水平与卵巢癌患者的恶性程度有关,上调LncRNA linc00261表达可抑制卵巢癌细胞SKOV3的增殖、迁移和侵袭能力,其可能成为卵巢癌早期诊断的潜在标志物和治疗新靶点。 Objective To investigate the expression of long-chain non-coding RNA(LncRNA)linc00261 in ovarian cancer and its potential mechanism in the development of ovarian cancer.Methods The expression of LncRNA linc00261 in ovarian cancer tissues and adjacent tissues of 60 patients with primary ovarian cancer was detected by qRT-PCR,and the correlation between the expression of LncRNA linc00261 and clinicopathological characteristics of ovarian cancer patients was analyzed.The expression plasmid and control plasmid of LncRNA linc00261 were designed and synthesized and transfected into SKOV3 cells.The expression of LncRNA linc00261 in transfected cells was detected by qRT-PCR.MTT assay was used to detect cell proliferation activity,scratch test was used to detect cell migration ability,and transwell chamber method was used to detect cell invasion ability.Results The results of qRT-PCR showed that the average relative expression of LncRNA linc00261 in ovarian cancer tissues was significantly lower than that in adjacent tissues,and the difference was statistically significant(t=5.153,P<0.05).The expression of LncRNA linc00261 in cancer tissue was not related to age,menopause and pathological type(P>0.05),but was related to the degree of tissue differentiation,clinical stage,lymph node metastasis and ascites(P<0.05).The results of qRT-PCR showed that the expression level of LncRNA linc00261 in the expression plasmid group was significantly higher than that in the control plasmid group and empty vector group(P<0.05).MTT assay showed that the proliferation ability of SKOV3 cells transfected with LncRNA linc00261 expression plasmid was significantly lower than that of control plasmid group and empty vector group(P<0.05).Scratch assay showed that the cell migration ability of LncRNA linc00261 expression plasmid group was significantly lower than that of control plasmid group and empty vector group(P<0.05).Invasion experiments showed that the number of transmembrane cells in the LncRNA linc00261 expression plasmid group was significantly smaller than that in the control plasmid group and empty vector group(P<0.05).Conclusion The expression of LncRNA linc00261 is down-regulated in ovarian cancer tissues,and its expression level is related to the malignant degree of ovarian cancer.Up-regulation of LncRNA linc00261 expression can inhibit the proliferation,migration and invasion of ovarian cancer cells,which may become a potential marker for early diagnosis and a new target for treatment of ovarian cancer.
