CD151介导内皮细胞血管生成参与腹主动脉瘤发病

CD151-mediated Endothelial Cell Angiogenesis Involves in Pathogenesis of Abdominal Aortic Aneurysm

摘要 目的研究CD151介导的内皮细胞血管生成在腹主动脉瘤发病机制中的作用。方法在Apoe基因敲除小鼠(Apoe-/-)皮下植入渗透性微泵建立血管紧张素Ⅱ诱导的小鼠腹主动脉瘤模型,免疫组化法检测动脉瘤组织CD151表达;在脐静脉内皮细胞(HUVECs)中转染CD151 siRNA或CD151重组腺相关病毒(rAAV-CD151),调控CD151表达,研究CD151对内皮细胞血管生成的影响;免疫荧光法检测动脉瘤组织CD31阳性细胞数以判断血管新生情况。结果①成功建立腹主动脉瘤小鼠模型;②在AngⅡ诱导的小鼠腹主动脉瘤模型动脉瘤组织内CD151表达升高;③CD151促进HUVECs血管生成;④腹主动脉瘤小鼠动脉瘤组织中血管生成增加。结论 CD151通过促进内皮细胞血管生成参与腹主动脉瘤发生发展过程。 Objective To study the physiopathologic role of CD151-mediated endothelial angiogenesis in abdominal aortic aneurysm(AAA).Methods The angiotensin Ⅱ-induced AAA model was established in Apoe-/- mice.The activation of CD151 was studied by immunohistochemistry.CD151 siRNA or CD151 adeno-associated virus was prepared and transfected into umbilical vein endothelial cells to detect angiogenesis.Immunofluorescence method was used to detect the expression of CD31,a marker of angiogenesis in AAA tissues.Results The mouse model of AAA was successfully established.The expression of CD151 was increased in Ang Ⅱ-induced AAA model.CD151 promoted angiogenesis of vascular endothelial cells and increased angiogenesis in abdominal aortic aneurysm mice.Conclusion CD151 may play a role in abdominal aortic aneurysm by promoting endothelial angiogenesis.
