操作台温度对小鼠卵母细胞受精及胚胎发育的影响

摘要 目的:探讨操作台温度对小鼠卵母细胞受精及胚胎发育的影响。方法:应用超促排卵技术获得小鼠卵母细胞,体外培养后受精,继续培养胚胎至囊胚期。观察在不同的操作台温度下小鼠卵母细胞的受精及胚胎的发育情况。结果:卵母细胞收集期操作台温度升高至38.0±0.2°C,两原核形成率(2PN率)和囊胚形成率分别为89.82%和41.66%,显著低于操作台温度为37.0±0.2°C的2PN率96.25%(P<0.05)和囊胚形成率57.50%(P<0.05)。胚胎培养期操作台温度升高至38.0±0.2°C,受精率、卵裂率和囊胚形成率与操作台温度为37.0±0.2°C时期的受精率、卵裂率和囊胚形成率无显著差异。结论:卵母细胞收集期操作台温度升高至38.0±0.2°C会降低小鼠卵母细胞2PN受精率、胚胎卵裂率和囊胚形成率,提高异常受精率。胚胎培养期操作台温度升高对小鼠卵母细胞受精率、胚胎卵裂率和囊胚形成率无影响。 Objective:To investigate the effect of different operating table temperature on fertilization and embryonic development of mouse oocytes.Methods:Mouse oocytes were obtained by hyper-ovulation and fertilized in vitro,then the embryos were cultured to blastocyst stage.The fertilization of mouse embryo oocytes and the development of the embryos were observed at different operating table temperatures.Results:When temperature of the operating table increased to 38.0±0.2°C,the pro-nucleate stage(2 PN)rate and blastocyst formation rate were 89.82% and 41.66%,respectively.Both rates were significantly lower than those when temperature was37.0±0.2°C,which were 96.25% and 57.50%,respectively(both P<0.05).There were no significant differences in fertilization rate,cleavage rate and blastocyst formation rate between the above mentioned stage temperatures at the stage during embryo culture period.Conclusion:Increasing the operating table temperature to 38.0 ±0.2° C during oocytes collection can decrease the fertilization rate of 2 PN,cleavage rate and blastocyst formation rate,and increase the abnormal fertilization rate.There were no effects on fertilization rate,cleavage rate and blastocyst formation rate when the temperature of operating table increased during embryo culture.
