摘要
为了揭示鸡肾型传染性支气管炎病毒 (IBV)免疫学特性 ,为生产上防治该病服务 ,应用 SDS- PAGE分析了 IBV的结构蛋白 ,发现肾型 IBV- X株与 M4 1和 H5 2的蛋白电泳图谱存在一定的相似性 ,均具有分子量为 90 0 0 0、84 0 0 0、6 70 0 0和 4 30 0 0的蛋白带 ,但 X株病毒还具有一条分子量为 6 10 0 0的条带 ,而且它的相对含量很高。肾型传支上海野毒 S株与 X株相似 ,也具有这条 6 10 0 0的条带 ,应用抗 IBV- X血清进行免疫转印 ,显示分子量 6 10 0 0的结构蛋白具有较强的免疫反应性。为了进一步揭示肾型 IBV诱导免疫保护的特性 ,应用 IBV- X毒株接种 10日龄鸡胚尿囊腔 ,收集尿囊液 ,灭活后 ,制成油乳剂灭活苗免疫鸡 ,30 d后 ,对 IBV- S毒株攻击的保护率达 10 0 % ,并呈现良好的体液免疫应答 ;疫苗在 4℃保存 7个月后用来免疫鸡 ,其保护率达95 %。该疫苗安全、可靠 。
IBV was multiplied and purified by ultracentrifugation, which proteins were electrophoretically seperated on 12 5% SDS polyacrylamide (SDS PAGE) gel, the results showed that IBV X, IBV M 41 and IBV H 52 had the similar profile with protein of 90 KD, 84 KD, 67 KD and 43 KD, protein of 61 KD with high relative content appeared in IBV X. A virulent strain of IBV isolated from Shanghai had the similar profile with IBV X, immunoblot analysis was performed with antibody against IBV X, the results suggested that the protein of 61 KD might to the protective antigen of IBV. In order to prevent this disease, the vaccine was prepared with formalized allantoic fluid. 220 chickens (1 month old) were immunized with the vaccine at different dose by muscle injection. One month later, 100% protection were induced after challenge with the same strain, an enhanced and prolonged antibody response was confirmed by hemagglutination inhibition test (HI). Moreover, the vaccine showed absolute safety and high stability.
