GDNF在脑胶质瘤中的表达及其与肿瘤恶性程度的关系 被引量:1

摘要 为探讨胶质细胞源性神经营养因子 (GDNF)在正常脑组织和恶性程度不同胶质瘤中的表达 ,及其与胶质瘤恶性生物学特性的关系 ,采用特异性抗体分别与 10例正常脑组织和 5 9例恶性程度不同的脑胶质瘤标本中存在的 GDNF结合 ,采用链霉菌抗生物素蛋白 -过氧化物酶连接法 (SABC)显色 ,观察 GDNF的表达情况。结果显示 ,GDNF在脑胶质瘤标本中的总阳性率显著高于正常脑组织 ,随着胶质瘤恶性程度增加 ,GDNF表达强度也逐渐增加 ,各级胶质细胞瘤差异显著。认为 GDNF可能是胶质瘤恶性增殖过程中的重要参与因子 ,可影响胶质瘤的发生、发展及其生物学特性。 GDNF的变化可作为胶质瘤恶性程度的判断指标 ,并可作为治疗胶质瘤的辅助手段。 To investigate the relation between glial cell line-derived neurotrophic factor(GDNF)and malignant biological activities by observing the expression of GDNF in normal brain tissue and human brain gliomas with different-malignancy.The specific anti-GDNF was binded GDNF existing in 10 cases of normal brain and 59 cases of human gliomas and then colored using SABC.Result showed the general positive rate of GDNF in gliomas was much higher than that in normal.With the increasing malignancy of gliomas,GDNF expressed more strongly,the differences were significant.This suggests GDNF maybe participate in the malignant proliferating of gliomas as an important factor to affect the kind of tumor's origination,development and other malignant biological activities,the level of GDNF may be a reliable standard to distinguish the malignancy of gliomas.Gliomas can be treated with the assistance of GDNF.
