摘要
目的 探讨多巴胺 (DA)诱导的PC12细胞凋亡是否与内源性一氧化氮 (NO)生成有关。 方法 酶还原法测定NO浓度 ,RT PCR法检测诱导型一氧化氮合酶 (iNOS)和caspase 3mRNA表达水平 ,免疫细胞化学方法和蛋白印迹法分别检测活化型caspase 3蛋白和iNOS蛋白水平。 结果 在多巴胺诱导PC12细胞凋亡过程中 ,iNOSmRMA和蛋白表达明显增加 ,内源性NO生成增多 ;caspase 3mRNA和活化型caspase 3蛋白的表达也明显增加。 结论 内源性NO可能参与多巴胺对PC12细胞凋亡的诱导作用。
Objective To study whether the endogenous nitric oxide (NO) was involved in dopamine (DA)-induced apoptosis in PC12 cells. Methods The level of NO was measured by the method of enzyme reduction. The levels of inducible nitric oxide synthetase(iNOS) and caspase-3 mRNA were determined by reverse transcription polymerase chain reaction(RT-PCR). Activated caspase-3 protein and iNOS was detected by immunocytochemistry and immunoblotting, respectively. Results Results During the development of apoptosis induced by DA, the levels of iNOS mRNA and NO increased markedly, as well as iNOS protein. Meanwhile, caspase-3 mRNA levels and activated caspase-3 protein were elevated compared with the control group. Conclusions Conclusions Endogenous NO may be involved in DA-induced apoptosis in PC12 cells.
