摘要
通过间歇性加药逐步提高培养液中阿霉素(ADM)的浓度,我们从人胃腺癌细胞SGC—7901中获得了耐ADM的细胞亚株。该耐药亚株对ADM具有25倍的耐受性,同时对其它多种抗癌药具有交叉耐受性。与亲代细胞相比,耐药细胞的某些生物学特性发生了改变:细胞内ADM的聚积量降低1倍;细胞内ADM的分布也发生了改变。该耐药亚株的获得,将为深入研究人胃癌耐药机制以及寻找克服耐药性的药物和方法,提供一个重要的体外实验模型。
An adriamycin (ADM) -resistant human gastric adenocarcinoma cellsubline SGC-7901/AR was obtained by intermittent stepwist selection in increasing concentration of drug from the parent cell line SGC- 7901. The resistant cells were 25 fold more resistant to ADM and displayed a multidrug-resistant phenotype. As compared to the parent cells,SGC-7901/AR cells had some remarkably altered biological properties;cellular content of ADM was reduced by approximately one half; the distribution of intracellular ADM had been changed. This resistant cells may be used as an experimental system for the study of mechanisms of drug-resistance and the measures for overcoming drug-resistance in human gastric cancer.
