摘要
将充分菌毛化的大肠杆菌C1976_79(pap +)经 4‰甲醛灭活作为免疫原免疫BALB/c小鼠 ,经淋巴细胞杂交瘤技术 ,用间接ELISA筛选获得FA1、FB11两株抗FB11菌毛单抗 ,其ELISA效价分别为 18log2 和 15log2 试管凝集价分别为 8log2 和 6log2 。免疫印迹实验表明 :两株单抗均能与鸡大肠杆菌F11菌毛反应 ,识别分子量为 18kD左右的菌毛蛋白。同时对猪病原性大肠杆菌进行检测 ,结果均无交叉反应。这表明F11菌毛是不同于K88、K99、987P、F4 1及F18等粘附素的一种大肠杆菌菌毛。
Escherichia coli C1976_79 posses F11 fimbriae,was inoculated BALB/c mice directly as immunogen to prepare specific monoclonal antibody.2strains of monoclonal antibody hybridoma cells were obtained by indirect Enzyme_linked Immunosrbent Assay(ELISA).All the 2 strains were specific to the F11 fimbriae through Western_blotting.When use the antibody as probe to check the fimbrias types with Escherichia coli.such as K88,K99,987P,F41 and F18.The result shows that the F11 fimbriae of pathogenic avian Escherichia coli are different with those adhesive factors.
