摘要
目的 :了解血透及不同透析膜对尿毒症患者IL - 8表达的影响。方法 :采用酶联免疫吸附试验 (ELISA)测定IL - 8血浆水平及测定PBMC培养上清液IL - 8水平。结果 :正常对照组、尿毒症非透析组及血透治疗组之间的IL - 8血浆水平均有显著性差异 ,聚砜膜组的IL - 8血浆水平低于铜仿膜组及铜氨膜组 ,而铜仿膜组及铜氨膜组之间无显著差异。血透患者透后的IL - 8血浆水平较血透前升高。血透患者的PBMC受LPS刺激后产生的IL - 8多于正常人及尿毒症未透析病人。结论 :血透可引起尿毒症患者IL - 8表达的异常 ,再生纤维素膜 (尤其是铜仿膜 )导致IL - 8血浆水平改变的能力强于聚砜膜 ,测量IL - 8表达有助于评价不同透析膜的生物相容性。
Objective:To investigate the influence of the hemodialysis and the different dialysis membranes on the interleukin-8 expression in the uremic patients.Method:The IL-8 of plasma concentration and supernate by the cultivated PBMC were measured with enzyme-linked immunoabsorbent assay ( ELISA ).Results:There was a significant difference among the plasma IL-8 level of the uremic patients on conservative therapy,that of the control group and that of the patients receiving hemodialysis. The IL-8 plasma level of the patients using the regenerated cellulose membrane (cuprophan CU, cuprammonium rayon CR) is higher than that of those using polysulfone ( PS ) membrane. The IL-8 plasma level was increased after hemodialysis.The production of IL-8 from PBMC of the patients underwent hemodialysis was more than that of normal subjects or uremic patients on conservative therapy.Conclusion:Hemodialysis can lead to the IL-8 abnormal expression in the uremic patients, it is more serious when the cellulose membrane (especially CU) is used. The measurement of IL-8 expression in PBMC is useful to evaluate the biocompatibility of dialysis membranes.
