摘要
目的 :建立一种便捷的检测系统性红斑狼疮 (SLE)患者血清抗双链DNA抗体的方法。方法 :在斑点免疫金银染色法 (dot-immungold -silverstaining ,Dot-IGSS)的基础上 ,缩短反应和洗涤时间 ,建立微量快速斑点免疫金银染色法 (Microvolume -Fast -Dot-IGSS ,MFDot-IGSS)并与Dot-IGSS进行比较。结果 :用MF Dot-IGSS和Dot-IGSS同步检测 70份SLE患者血清抗双链DNA抗体 (A -dsDNA) ,阳性率分别 6 4 .3%和6 5 .7% ;检测其他自身免疫性疾病患者血清 30份阳性率均为 3.3% ;检测健康献血员血清 5 0份 ,阳性率均为2 .0 %。二种方法的敏感性和特异性均无显著性差异 (P >0 .0 5 )。结论 :MFDot -IGSS检测SLE病人血清A-dsDNA的敏感性和特异性与Dot -IGSS相似 ,但便简快速 ,适用于门诊及基层医院。
Objective:To set up a simple and convenient immunoassay for the diagnosis of SLE.Methods:A microvolume fast dot-immunogold silver staining(MFDot-IGSS) technique was set up on the basis of the conventional Dot-IGSS by shorting the incubation and washing time. The results obtained were compared with those of the conventional Dot-IGSS simultaneously.Results:The positive rates of the anti-dsDNA antibodies from patients with SLE were 64.3% by MF Dot-IGSS and 65.7% by Dot-IGSS respectively. The positive rates of the anti-dsDNA antibodies were both 3.3% from 30 cases with other autoimmune diseases (AID) and both 2.0% from 50 healthy controls by the two methods respectively. No significant differences were found in the sensitivity and specificity between the fast and the conventional assays ( P >0.05).Conclusions:The sensitivity and specificity of MF Dot-IGSS were similar to those of the conventional Dot-IGSS while the former is simple, timesaving and suitable for the basical hospital.
