摘要 建立了用单一工具酶 (CK)测定红细胞中三磷酸腺苷 (ATP)含量的比色测定法。在 PH9.0环境下 ,以肌酸激酶 (CK)催化肌酸与标本中 ATP反应 ,产生的磷酸肌酸用磷钼酸比色法进行检测 ,从而获得 ATP含量。线性范围0 .1~ 0 .9mmol/ g Hb,批内、批间平均变异系数分别为 1.6 4 %、2 .12 % ,平均回收率为 10 1.8% ,与己糖激酶法有极好的相关性 (r=0 .999,y=4 0 .89+1.0 0 6 x)。结果表明本法具有良好的精确性 。 A single method for the determination of ATP content in blood red cell was introduced. CPK could catalyze the reaction both creatine and ATP in sample while pH was 9.0, then the phosphomolybidic acid by means of the colorimetric analysis. The amount of phosphocreatine produced was proportional to the ATP contect. The linerity scope was from 0.1 mmol to 0.9 mmol, and the average coefficient of variation of within day and day to day were 1.64 % and 2.12 % respectively.The average recovery rate was 101.8 %, and there was a good correlation when compared with the method using HK system (r=0.999,y=40.89+1.006x). The results shows that single enzyme and ordinary colorimeter was not only a good accuracy but also practicality methed.
