恶性疟原虫DNA疫苗在小鼠体内组织分布和安全性的初步研究 被引量:1

摘要 目的 探讨疟疾DNA疫苗在小鼠体内的组织分布 ,并对其安全性进行观察。方法 将重组质粒pBK -CSP经肌肉途径免疫BALB/c小鼠 ,分别在 4周和 8周后剖杀动物并摘取各种组织 ,抽提全组织DNA进行PCR扩增后经琼脂糖凝胶电泳分析 ,并对DNA疫苗的安全性进行观察。结果 免疫 4周和 8周后 ,仅有DNA疫苗接种位点的肌肉组织检测到CSP基因 ,而 10 0 μg的质粒DNA并未产生明显的毒副作用。 结论 疟疾DNA疫苗接种 4周后 ,质粒DNA仅分布于接种部位 ,并可持续至 8周以上 ,而未发现毒副作用。 Aim To explore the tissue distribution and safety in mice immunized with DNA vaccine (pBK-CSP),a recombinant plasmid encoding gene of the malaria circumsporozoite protein from Plasmodium falciparum (PfCSP) Methods The recombinant plasmid pBK-CSP was injected into BALB/c mice by intramuscular route Various tissues were removed from mice at 4 and 8 weeks postinjection Total tissue DNAs were extracted for PCR amplication The results were identified by agarose gel electrophoresis The safty of DNA vaccine were surveyed Results CSP gene fragments were amplified only in the muscular tissues of injection site at 4 and 8 wk postinjection In safety stuty,plasmid DNA at dose of 100 μg had no obvious adverse effects Conclusion Except for the site of administration,plasmid DNA is no longer detectable in tissues after 4 wk postadministration
