摘要
目的 观察蒿甲醚 (Art)对日本血吸虫的脂质过氧化损伤。方法 小鼠感染血吸虫尾蚴 5周后 ,1次灌服Art30 0mg/kg ,于治疗后 6~ 2 4h剖杀 ,收集雄虫 ,用硫代巴比妥酸 (TBA)比色方法测定其TBA反应物 (TBARs)水平。并取未经药物治疗的合抱虫 ,置于含Art和 /或氯化血红素 (hemin)的培养液内培养 2 4h后 ,作TBARs分析。结果 小鼠体内的血吸虫雄虫经Art作用后 12和 2 4h ,其脂质过氧化物水平均有明显升高。体外 ,Art或hemin单一作用血吸虫 2 4h ,对其TBARs水平无影响 ,但Art(15~ 6 0 μmol/L)与hemin(10~ 5 0 μmol/L)伍用可导致虫体的TBARs水平明显升高。结论Art具有血红素依赖的诱发日本血吸虫脂质过氧化作用。
Aim To study artemether(Art) induced lipid peroxidation in Schistosoma japonicum Methods In vitro,thiobarbituric acid reactive substances(TBARs),the indicator of lipid peroxidation,were detected in five week old paired worms incubated without or with Art and/or hemin for 24 h In vivo,TBARs were detected in male worms freshly recovered from mice 6~24 h after treatment with Art 300mg/kg Results Though Art or hemin alone had no effect on the basal TBARs levels,a combination of Art(15~60μmol/L)and hemin(10~50μmol/L)led to a significant TBARs increase in the parasite in vitro The drug effect on the parasite demonstrated in vivo After male worms were exposed to Art for 12~24 h in vivo,the TBARs levels were significantly increased in a time related pattern Conclusion Art showed a heme dependent inducement of lipid peroxidation in schistosomes
