摘要
通过使用混合激素诱发籼稻871015、871109花粉愈伤形成胚性细胞;对籼稻花粉胚性细胞团和粳稻绿芽丛进行分割培养,快速繁育大群体绿苗获得成功。籼稻两个组合的8块胚性细胞团,在两个月内经3—5次切割培养获得绿苗12690株;粳稻花培绿芽丛仅经一次分割培养后的净增绿苗率127.2%,大幅度地提高了水稻花药培养再生绿苗率。分割培养的再生苗,健壮的形成特征:苗粗、浓绿、根多、洁白。良好的生理素质:发根力强,平均每棵试管苗多增根1.8条,根长度增长168.4%;平均每棵再生苗鲜重和干重分别增加0.29克和0.175毫克。
Embryonic cells were induced and the clone of them in anther culture in indica rice was successively established, and further a great number of green plantlets by way of cell embryogenesis were obtained. Low concentration of hormones (6—BA0.5mg/L+IAA0.25mg/L+KT 0. 5mg/L)was favorable to the form of embryonic cell from the pollen callus of indica rice 871015 and 871109. These embryonic cell grew and produced embroes which can regenerate plantlet. Within two months, 12690 green plantlets were regenerated from eight embryonic cell masses which had been cut and cultured 3- 5 times repeatedly. The embryonic cell mass also produced albino plantlets. Though the frequency of plantlet (green meristemoid) regenerated from the clone of the embryonic cell in subculture for one year was as high as ninety per cent, the frequence of distortion was also high and almost none of them transplanted in soil can be alive. Chromosome of embryonic cells in subculture for one year varied widely. Chromosomal number obviously increased. Of 532 cells inspected cells with n>12 made up 83.08%, 54.89% of which had. 18—20 chromosomes. However, they containea a set of complete haploidy chromosome at least.
