长白山野生黄蘑抗肿瘤活性多糖的研究 被引量:13

ANTITUMOR ACTIVITY OF SOME POLYSACCHARIDESISOLATED FROM THE WILD MUSHROOM" HUANGMO" FROM MT. CHANGBAI

摘要 用热水,1%草酸铵和5%氢氧化钠依次抽提出长白山野生黄蘑子实体中的多糖组分FⅠ,FⅡ和FⅢ。再经乙醇沉淀、离子交换层析、凝胶过滤和亲和层析等方法进一步分离纯化,得到20余种多糖成份。通过皮下注射小鼠Sarcoma 180实验法对各多糖组分进行抗肿瘤活性检定,FⅠ_0,FA—1,FA—2,FⅡ—2,FⅢ—1—b和FⅢ—2—b等6组份具有较强的抗肿瘤活性。其中初步判定结果;FⅠ_0为异半乳聚糖和β—D—葡聚糖的混合物;FA—1及FA—2为含有异半乳糖和蛋白质的β—D—葡聚糖;FⅡ—2为β—D—葡聚糖;FⅢ—1—b和FⅢ—2—b为β—1,3;1,6—D—葡聚糖。 Polysaccharides were extracted from a Chinese mushroom, ' Huangmo' with hot water (F Ⅰ ), 1 % ammonium oxalate solution (F Ⅱ ), and 5 % sodium hydroxide solution (F Ⅲ), and further 20 polysaccharides were isolated and prepared by ethanol precipitation, ion -exchange chro-matography, gel filtration and affinity chrqmatography. Of the polysaccharides obtained, six fractions, F Ⅰ 0, FA-1, FA -2, F Ⅱ -2, F Ⅲ -1 -b and F Ⅲ -2 -b, were selected on the basis of the results of the antitumor activity test (the Sarcoma 180/mice, i. p. method). Six fractions which showed relatively high activity were identified; F Ⅰ 0, as a mixture of heterogalactan and β-D-glucan; FA - 1 and -2, as β-D-glucan accompanied with heterogalactan and protein; F Ⅱ -2, as β-D-glucan; FⅢ -1-b and -2 -b, as (1→3) ; (1→6) -β-D-glucan.
