河南食管癌高发区食管癌组织中COX-2的表达

Expression of cyclooxygenase-2 protein in human esophageal carcinoma tissues on the subjects at high-incidence area for esophageal cancer in Henan

目的 :探讨河南食管癌高发区食管癌组织中COX 2表达变化与食管癌细胞分化程度及淋巴结转移的关系。方法 :免疫组化ABC方法检测 31例食管鳞癌组织中COX 2的表达 ,以活检正常食管粘膜 10例为对照。结果 :COX 2免疫阳性反应主要定位于食管癌细胞的细胞质 ,正常对照组无COX 2表达。 31例食管癌组织中有 16例 (5 1.6 % )表达COX 2 ;食管癌组织分化的恶性程度与COX 2免疫反应的强度呈负相关 (r =- 0 .5 6 4 ;P =0 .0 0 1) ;伴淋巴结转移者 6 1.9%COX 2阳性 (13/2 1) ,但统计分析未发现COX 2的免疫反应程度与有无淋巴结转移间存在相关性 (K SZ =0 .830 ;P =0 .4 96 )。结论 :COX 2过度表达可能与食管癌细胞分化程度有关 ,可能成为食管癌防治 ,特别是高分化食管癌治疗的新靶点。

Aim: To characterize COX 2 expression in esophageal cancer tissue and to correlate the alterations of COX 2 expression with esophageal cancer cell differentiation and lymph node metastasis.Methods: Immunohistochemistry method (ABC) was applied to determine the expression of COX 2 in 31 surgically resected human esophageal cancer specimens. Normal esophageal mucosa biopsy ( n =10) was used as control. Results: Immunoreactivity of COX 2 was chiefly located in the cytoplasm of esophageal caner cells. No positive immunoreactivity was observed in control group. The positive immunostaining rate for COX 2 in 31 esophageal cancer patients was 51.6% (16/31). The differentiation progression of esophageal carcinoma was inversely related to the expression of COX 2 by Spearman rank correlation test ( r =-0.564; P =0.001), but there was no correlation between metastasis characteristics of esophageal cancer and intensity of COX 2 protein expression ( Z =0.830; P =0.496).Conclusion: COX 2 overexpression may be one of the key molecular events correlated to the differentiation of esophageal cancer, and it further provides a new research target for esophageal cancer prevention and treatment.
