摘要
目的探讨前纤维蛋白在增生性瘢痕形成与收缩中所起的作用及意义。方法收集患者不同时期增生与非增生瘢痕作为研究对象,利用前纤维蛋白的基因特异片段,制成寡核苷酸探针与瘢痕组织切片原位杂交,同时,将各标本进行组织块培养,制成成纤维细胞爬片,进行原位杂交,分析基因在组织与细胞中变化规律。结果前纤维蛋白基因在早期增生性瘢痕中表达均明显强于晚期增生及非增生瘢痕。结论瘢痕增生挛缩与细胞骨架运动的相关基因存在密切关系,而前纤维蛋白起到十分重要的作用。同时,如果抑制该基因的表达,可能实现减轻瘢痕挛缩。
Objective To investigate the effect of gene exp ression of fibroblast profilin on th e contraction of hypertrophic scar.Method profilin,one of the most important g enes in cytoskeleton and movement of hypertrophic scar,was selected and made into oligonucleotide probe.24cases of hypertrophic scar and 24non-hypertrophic scar and 12n ormal skin were used and these scar we re taken on 3,6,9,12months after burned.Freezing slice s and cultured fibroblasts were made to detect the expression of these gen es by hybridization in situ.Results profilin were found the expression i n scar tissue,but those in hypertrop hic scar were significantly stronge r than non-hypertrophic scar.Conclusions Over-expression of cytoskeletal re lative genes of profilin act very imp ortant functions on the contraction of hypertrophic scar.
