摘要
在缓冲液中 ,利用 1 β D 呋喃核糖基 1,2 ,4 三唑 3 酰胺 (ribavirin)和 1 (2′ 脱氧 β D 呋喃核糖基 ) 1,2 ,4 三唑 3 酰胺 (deoxyribavirin)作为配体 ,分别与cis [Pt(DMSO) 2 Cl2 ]或K[Pt(DMSO)Cl3 ]进行配位反应 ,得到了高产率的 [Pt (N4,N7 ribavirin) (DMSO)Cl](1)和 [Pt(N4,N7 deoxyribavirin) (DMSO)Cl](2 )两种配合物 .1 (2′ 脱氧 β D 呋喃核糖基 ) 1,2 ,4 三唑 3 酰胺的合成是由 1 β D 呋喃核糖基 1,2 ,4 三唑 3 酰胺与 1,3 二氯 1,1,3,3 四异丙基二硅氧烷保护 ,在 1 甲基咪唑存在下与硫代苯氧基碳酰氯反应 ,再用tri n butyltinhydride和AIBN还原 ,四丁基氟化铵的四氢呋喃溶液脱去保护基四步反应完成的 .
Platinum(Ⅱ) complexes of [Pt(N 4,N 7 ribavirin)(DMSO)Cl] and [Pt(N 4, N 7 deoxyribavirin) (DMSO)Cl] were prepared by the reactions of cis [Pt(DMSO) 2Cl 2] or K[Pt (DMSO)Cl 3] with 1 β D ribofuranosyl 1,2,4 triazole 3 carboxamide and its deoxyanalogue in buffer with good yields. The preparation of 1 (2′ deoxy β D ribofuranosyl) 1,2,4 triazole 3 carboxamide was also performed through a four step procedure, including protection of 3′,5′ dihydroxyl group of ribavirin with 1,3 dichloro 1,1,3,3 tetraisopropyl disiloxane, phenoxythiocarbonylation of the 2′ hydroxyl group of 3′,5′ O TPDS ribavirin with phenoxythiocarbonylchloride, reduction of 2′ O phenoxythiocarbonyl ester of 3′,5′ O TPDS ribavirin with tri n butyltin hydride and AIBN, and deprotection of 3′,5′ O TPDS ribavirin with tetrabutylammonium fluoride in THF.
