摘要
本文对外径1.0~1.6mm(平均1.41±0.17mm)家兔颌外静脉进行快速冷冻保存和两步冷冻保存的比较研究,并观察其作为同种异体移植物修复静脉缺损的通畅性。结果表明,快速冷冻保存的静脉,内膜损伤严重,移植后1~12周内,血栓形成率为85%,而两步冷冻保存的静脉,内膜损伤较轻,移植后1~12周,通畅率为70%。文中对冷冻损伤的发生机制及预防措施进行了讨论,强调保持内膜完整性是提高静脉移植通畅率的重要因素。
Rabbit veins with an external diameter of 1. 0~1. 6 mm (mean 1. 41±0. 17mm) were inves-gated in vitro either after preservation by cooling rapidly to - 196℃ or after storage in liquid nitrogen by a two-step freezing schedule,and the patency rate of the veins as allografts interposed between defects of femoral veins was observed;the results showed that a striking intimal damage was seen in the veins plunged directly into liquid nitrogen with a very high rate of thrombosis (85%),while the light microscope showed an intact endothelium in the veins frozen by a two-step procedure, which probably accounted for a long-term patency rate of 70% at 3 months. The mechanisms of cryoinjury and cryoprotection are discussed with special emphasis on the contributing factor of the preserved endothelium for the maintenance of long-term patency.
