摘要
目的 :探讨显微操作法对获取孕妇外周血中胎儿有核红细胞 (NRBCs)的可行性和准确性。方法 :将妊娠 6~ 38周 4 1例孕妇的外周血进行不连续密度梯度离心 ,将分离后的细胞进行制片 ,在光学显微镜下识别NRBCs,然后用微操作仪获取NRBCs,并用聚合酶链反应 (PCR)在基因水平进行验证。结果 :4 1例孕妇有 2 2例在外周血中检出NRBCs ,检出率为 5 3.7% ,PCR结果显示 2 2例中有 10例呈现Y特异带 ,总符合率为 95 .4 %。结论
Objective:To study the effectiveness of isolating fetal nucleated red blood cells(NRBCs) using a micromanipulator.Method:We studied 41 women of 6~38 weeks' gestation. Fetal cells were isolated from maternal blood with discontinuous density gradient centrifugation. NRBCs were found and retrieved using a micromanipulator under a microscope. The NRBCs were analysed by polymerase chain reaction(PCR) amplification to determine the presense of a Y chromosome-specific repeat sequence in mothers carrying male fetuses.Result:22 cases of 41 cases were detected NRBCs, The ratios of NRBC detection was 53.7 % .PCR showed 10 cases Y specific DNA sequence were dectected in 22 cases.Conclusion:Micromanipulator is a correctly unique method in isolating NRBC from maternal blood.
