摘要
目的:研究紫杉醇对肿瘤细胞增殖及基质金属蛋白酶(MMP-2,MMP-9)表达的影响。方法:半定量RT-PCR法检测紫杉醇对体外培养的HO8910卵巢癌细胞MMP-2和MMP-9基因mRNA表达的作用。结果:(1)MTT法检测提示紫杉醇在浓度达100 nmol·L^(-1)以上时,对HO8910细胞生长抑制率>30%(P<0.001),并随浓度增加而升高(r=0.6574,P<0.01);(2)紫杉醇浓度在10 nmol·L^(-1)时MMP-2和MMP-9 mRNA表达降低(P<0.01),其改变呈剂量依赖性下降。结论:紫杉醇在抑制HO8910细胞增殖时,也抑制基质金属蛋白酶MMP-2和MMP-9mRNA的表达,因而,可能产生抑制肿瘤增殖和侵袭转移的作用。
Objective: To investigate the effects of paclitaxel acting on matrix metalloproteinases ( MMP - 2, MMP - 9 ) expression by cancer cells. Method: It were assayed the expression of MMP - 2 mRNA and MMP - 9 mRNA in overian cancer H08910 cell treated by paclitaxel in vitro by RT - PCR. In semi - ratio. Results: (1 ) It was suggested that the growth inhibitive rates were higher than 30% while concentration of paclitaxel were above 100 nmol · L-1 assayed by MTT method (P <0. 001) , and the rate increased significant correlation with raising the concentration of the agent (P <0. 01 ) . (2) The expression of MMP - 2 and MMP -9 were down - regulated by paclitaxel consistently at 10 nmol · L-1 investigated with RT - PCR, and the expression of MMP -2 mRNA decreased following raising dosages as well as MMP -9. Conclusion: Paclitaxel restrained proliferation of H08910 ovarian cancer cell, and inhibited meanwhile the expressions of MMP -2 mRNA and MMP -9 mRNA in the cell. Therefore, it may be possess the effects of choking back proliferation and invasive metastasis of cancer cell.
