氯沙坦对STZ糖尿病大鼠肾脏结构和功能的影响

THE EFFECTS OF LOSARTAN ON RENAL STRUCTURE AND FUNCTION IN STZ DIABETIC RATS

目的 :观察血管紧张素受体拮抗剂氯沙坦对糖尿病大鼠肾脏功能和肾组织结构的影响。方法 :雄性Wis tar大鼠分为正常对照组 (C)、糖尿病对照组 (D)、糖尿病氯沙坦治疗组 (DL)。于实验第 6、12和 2 4周 ,测定各组大鼠的血糖、糖化血红蛋白、肌酐清除率及尿蛋白排泄率 ,并对肾脏标本进行了光镜、电镜观察 ,同时测量各组大鼠的平均肾小球体积。结果 :于第 6周、12周、2 4周 ,糖尿病对照组大鼠的上述指标与C组相比 ,均明显升高 (P <0 .0 5 ) ,而DL组尿蛋白排泄率均低于同时期的D组 (P <0 .0 5 )。DL组肾小球体积显著低于D组(P <0 .0 5 )。电镜检查发现D组基底膜显著增厚 ,系膜和系膜细胞显著增生 ,DL组上述改变明显减轻。结论 :氯沙坦能够延缓糖尿病大鼠肾脏病变的进程 ,保护肾功能。

Objective:To observe the effects of osartan on renal structure and function in diabetic rats.Methods:Male Wistar rats were randomly divided into three groups; There were normal control group(C group),streptozotocin (STZ) induced diabetic rats group (D group) and diabetic rats treated with Losartan group (DL group). Blood glucose, glycosylation creatinine clearance and urinary protein excretion of rats were measured in 8,12 and 24 weeks,the kidneys were processed for morphological examination with light and electron microscopes. Mean glomemlar volume(MGA) was calculated using stereological method.Results:In 8,12,and 24 weeks, The body weight, blood glucose and GHbA1 in STZ-induced diabetic rats were significant higher than that in C groups. The urinary protein excretion were lower than that in C group( P <0.05 ), in DL group glomerular volume(MGV) in DL group was significant smaller than that in D group ( P <0.05). The electron microscopic results showed significant mesan gial expansion, glomemlar basement membrane(GBM) thickening in D group, but abnormal changes were alleviated to a certain extent in DL group.Conclusions:Losartan can prevent renal pathological progress in diabetic rats. It is suggested that losartan may have some renal protective effects.
