摘要
利用 RT-PCR及序列测定对 5株猪瘟野毒 1 4个不同代次毒株 E2基因主要抗原编码区序列进行了分析。结果表明每个野毒的不同代次毒株核苷酸及氨基酸序列均呈现较高的保守性 ,核苷酸及氨基酸序列均无变化 ,核苷酸及氨基酸同源性均为 1 0 0 %。与 0 2号野毒相比 ,其他 4株野毒核苷酸及氨基酸同源性分别为 82 .0 %~ 99.5%、84.1 %~ 98.6 %。遗传衍化分析结果表明所有毒株被分成 2个基因组 ,每个基因组又分成 2个基因亚组。 0 3、0 5和 2 2号毒株位于第一基因组 ,0 2、0 6号毒株位于第二基因组。每一个毒株的不同代次毒株均位于同一基因组、同一基因亚组。
The major coding sequence of E2 of different generations of HCV field strains were analysed by reverse trancriptase polymerse chain reation (RT PCR) and sequenced.The results showed that the nucleotide sequence and amino acid sequence of the major coding sequence of E2 gene of differents generation of every field strain were conservative. The nucleotide sequence and amino acid sequence of all generation of every strain were not change.The identity of nucleotide sequence and amino acid sequence were 100%.Compared with the 02 strain,the identity of nucleotide sequence and amino acid sequence of the others was 82.0%~99.5%,84.1%~98.6%. The phylogenetic tree was divided into 2 groups,per group was divided into 2 subgroups.03,05 and 22 strains were distributed in 1 group.02 and 06 strains were divided into the 2 subgroup.Different generations of every strain were all divided into the same group,sama subgroup. It confirmed initinally that molecular structure of hog cholera virus was genetically stable.
