摘要
目的 建立血清中氟西汀的高效液相色谱检测方法 ,监测氟西汀血药浓度 ,为临床用药提供依据。方法 0 .5mL样品用 2 %异丙醇的正庚烷 5mL提取液萃取净化。色谱条件 ,以C18柱 (4μm ,15 0mm× 3.9mm)作为分析柱 ,流动相为甲醇 磷酸氢二铵缓冲液 (70∶30 ) ,pH为 6 .4 ,流速为 0 .6mL·min-1,柱温 4 0℃ ,检测波长 2 30nm。结果 氟西汀在 0 .3~ 6 μmol·L-1范围内呈线性 ,回收率为 98.8%~ 10 1.4 % (n =5 )。结论 本法简便、快速、准确 。
OBJECTTVE: To develop a HPLC method for the determination of fluoxetine in serum and provide a scientific basis for clinical use of fluoxetine. METHODS: 0.5 mL serum sample was used. Doxepin hydrochloride was used as the internal standard. It was then extracted by 5 mL n-heptane containing 2% iso-proalcohol, C18 column (4 μm, 150 mm × 3.9 mm) was used with the mobile phase of methanol sodium phosphate dibasic buffer solution (70:30). The flow rate was 0.6 mL·min-1 and the column temparature was 40°C. The detection wavelength was 230 nm. RESULTS: The linearity was obtained over the range of 0.3∼6μmol·L-1. The lowest detection limit of fluoxetine in serum was 60 nmol·L-1. The recoveries of three concentrations of fluoxetine in serum were 100.01%, 99.93 % and 99.93% respectively. CONCLUSION: This report provided a rapid, convenient, sensitive and accurate method which is suitable for monitoring the concentrations of fluoxetine in serum and its pharmacokinetic study.
