痹肿消汤对实验性关节炎大鼠血浆TNF-α的影响

Influence of bizhongxiao (bzx) decoction on the plasma TNF-α levels of CⅡ-induced rheumatoid arthritis in rats

目的 :观察痹肿消汤对实验性关节炎大鼠的血浆TNF α的影响 ,探讨痹肿消汤治疗类风湿性关节炎的作用机制。方法 :(1) 75只SD大鼠随机分为 4组 ,皮下注射Ⅱ胶原诱导实验性关节炎模型 ,采用放射免疫法检测各组不同时间大鼠血浆TNF α水平。结果 :造模大鼠 88.5 7%出现关节炎症状 ;造模 2 5d后 ,模型组、痹肿消汤组及甲氨喋呤组TNF α水平明显高于正常组 (P <0 .0 1) ;且模型组的TNF α水平明显高于痹肿消汤组及甲氨喋呤组 (P <0 .0 1) ;随着时间的延长 ,模型对照组TNF α逐渐升高 ,痹肿消汤组及甲氨喋呤组则逐渐降低 ;而痹肿消汤组TNF α水平低于甲氨喋呤组 (P <0 .0 5 )。结论 :TNF α在RA疾病滑膜组织炎症形成和发展过程中发挥着重要作用 ;痹肿消汤能下调血浆TNF α的水平 。

Objective To observe the influence of BZX decoction on the plasma TNF α levels of CⅡ induced arthritis (CIA) in rats. To explor the mechanism of BZX decoction in the treatment of rheumatoid arthritis disease. Methods Seventy five SD rats were randomly divided into four groups.The rat experiment arthritis model was established by subcutaneous injection ofⅡcollogen. The plasma TNF α levels of the four groups were tested with the radioimmunologic assay in different times. Results The incidence of arthritis in the rats immunized with CⅡ was approximately 88.57%. The plasma TNF α levels of the model group, BZX decoction group and methotrexate (MTX) group were notably higher than those of the normal group (P<0.01). The plasma TNF α levels of the model group were notably higher than those of the MTX control group and BZX decoction experiment group in different times (P<0.01). The plasma TNF α levels of the model group rose step by step,but those of the BZX decoction group and MTX group decreased. The plasma TNF α levels of the BZX decoction group were significantly lower than those of the MTX group (P<0.05). Conclusion TNF α plays a very important role in the formation and development of RA disease. BZX decoction can decrease the plasma TNF α levels. The effects of BZX decoction are significantly better than those of the MTX group.
