摘要
目的 :探讨Lac启动子在可溶性抗体Fab表达中的作用。方法 :利用DNA重组的方法对P3 -3G9噬菌体抗体表达载体[1 ] 进行改建 ,通过免疫印迹试验鉴定新的载体P3G9-L是否有可溶性Fab的表达。结果 :1.P3G9-L为单Lac启动子同时驱动Fd段及K链的双顺反子的可溶性Fab段表达载体。 2 .P3G9-L中的Lac启动子的作用是缓和的 ,可溶性Fab表达量较低。结论 :1 Lac启动于可以用于可溶性抗体Fab的表达体系。 2
Objective:To study the action of Lac promotor in the expression of antibody Fab.Methods:We tried to convert P3-3G9 in DNA recombination.The expression of soluable Fab fragment was proved by Western-blotting.Results:P3G9-L can express soluable Fab fragment and the function of Lac promotor is weak.The expression level is lower.Conclusion:Lac promotor can be used to express soluable Fab fragment.Double Lac promotor can be replaced by single Lac promotor in expression of Fab antibody.
