摘要 目的 :比较重组葡激酶 (r Sa K)不同给药途径治疗幼猪急性脑栓塞的疗效。方法 :采用颈内动脉注入体外血栓法建立幼猪急性脑栓塞动物模型 ,4小时后自颈部动脉导管或腹壁静脉应用 r Sa K(剂量分别为0 .8× 10 4 、1.6× 10 4 、3.2× 10 4 U / kg) ;对照组用安慰剂。治疗结束后 30、6 0和 90分钟行脑血管造影 ,同时检测凝血指标。结果 :动脉给药组和静脉给药组的血管再通率均高于对照组 (P均 <0 .0 5 ) ,但 2组之间无显著性差异 ;2组对凝血功能的影响均较小。结论 :颈内动脉或周围静脉应用 r Sa K均能安全、有效地溶化猪脑血栓。 Objective:To evaluate the thrombolytic efficacy and safety of recombinant staphylokinase (rSaK) in a pig model by different administration routes (intravenous versus regional intra arterial). Methods: An embolic pig stroke model was established by injecting fragments of human plasma clot into one internal carotid artery.Four hours after embolization,the pigs were blindly treated with rSaK by regional intra arterial or intravenous infusion in a dose of 0 8×10 4 U/kg or 1 6×10 4 U/kg or 3 2×10 4 U/kg.Angiographic efficacy and hemastatic function parameters were observed.Results:Both intraarterial and intravenous groups resulted in significant thrombus dissolution(all P <0 05),the difference in reopening rate of obstructed vessels between intraarterial and intravenous groups was not significant.The result of hemastatic function parameters showed no significant difference between both administration routes.Conclusions:The two modes of administration could effectively dissolve thrombus in the pig model of embolic stroke with similar efficacy.
