胃癌组织中诱导型一氧化氮合酶表达及其与增殖细胞核抗原、p53表达的相关性研究 被引量:3

摘要 目的:探讨胃癌组织中诱导型一氧化氮合酶表达,及其与增殖细胞核抗原(PCNA)、肿瘤抑制基因p53表达的相关性。方法:应用免疫组织化学SABC法检测85例胃癌组织、30例癌旁组织及25例正常胃黏膜组织中诱导型一氧化氮合酶(iNOS)蛋白表达,同时检测胃癌组织中PCNA、p53的表达。结果:81.2%胃癌组织中表达iNOS蛋白,而癌旁组织及正常胃黏膜组织则分别为33.3%和20.0%(P<0.001)。iNOS表达与胃癌组织的分化程度、临床分期及淋巴结转移有明显相关性,而与胃癌组织的大体类型、病理组织类型无明显相关关系。p53、PCNA表达与iNOS蛋白表达显著相关(P<0.05),而且iNOS表达阳性区,PCNA的阳性率高,占83%。结论:iNOS在胃癌组织中表达上调,提示iNOS可能在胃黏膜细胞癌变过程中起重要作用。iNOS表达可促进胃癌细胞增殖。iNOS的异常表达及抑癌基因p53的失活在胃癌的癌变过程中起协同作用。 Objective:To study the expression of iNOS and its relationship with the expression of p53?PCNA in human gastric carcinoma. Methods:Immunohistochemical assay was to detect the expression of iNOS?PCNA and p53 proteins in 85 samples of human gstric carcinoma, 30 samples of adjacent normal gastric mucous tissue and 25 samples of normal gastric mucous tissue. Results:Expression of iNOS protein was more detected in human gastric carcinoma (81.2%) than adjacent normal gastric mucous tissue (33.3%) and normal gastric mucous tissue(20 0%)(P< 0.001). There was relationship between iNOS protein expression and differentiation?clinical stages or lymph node metastases of carcinoma, but did not show significant difference among the groups with different pathological types and types of naked eye. The expression of iNOS was significantly correlated with p53 or PCNA expression (P< 0.05). The positive immunostaining rate for PCNA in the area of a positive immunostaining reaction of iNOS was high (83%). Conclusion:The up regulation expression of iNOS in human gastric carcinoma suggested that iNOS plays a role in the pathway of carcinogenesis in gastric carcinoma. The expression of iNOS may promote the proliferation of tumor cells. Inactivation of antioncogene p53 and up regulation of iNOS might play synergetic roles in the process of carcinogenesis of gastric carcinoma.
