摘要
目的 研究脂质体介导ANG反义核酸转染A5 49细胞后 ,对其中ANG蛋白表达的影响 ,探讨其作为抗血管生成与抗肿瘤药物的可行性。方法 用人工合成的人ANG反义寡聚核苷酸 ,以脂质体为载体转染人肺癌A5 49细胞 ,研究A5 49细胞在基因转染后 ,其ANG蛋白表达的变化。结果 脂质体转染程序优化后 ,得到最佳转染效率 ;免疫组化显示细胞内ANG蛋白表达明显减少 ;培养基ELISA检测显示每细胞分泌的ANG蛋白量与对照组相比有显著差异。结论 ANG反义寡聚核苷酸抑制A5 49细胞中ANG的表达 。
Objective To investigate the effects of ANG antisense oligonucleotide transfected by liposome on A549 cells and to explore the possibility as anti angiogenetic and anti tumor drugs. Methods Human lung cancer cell line A549 was transfected with a synthetic oligonucleotide. The changes of ANG protein oression was observed after transfection by cationic liposome. Results The liposome could effectively deliver ANG antisense oligonucleotide to A549 cells. When the cells were treated with antisense oligonucleotide, ANG protein level decreased greatly both in the cells and in the media. Conclusion Antisense ANG oligonucleotide can inhibit ANG expression specifically in A549 cells in vitro , providing the basis for further studies in vivo .
