Bcl-2及Bax在db/db自发性糖尿病小鼠颌下腺的表达研究 被引量:12

摘要 目的 观察转基因糖尿病小鼠颌下腺的形态学改变以及凋亡相关基因Bcl- 2和Bax在颌下腺表达的变化。方法 取 3、4、6、8、10月龄db/db糖尿病小鼠及相应月龄的db/ +m小鼠 (对照组 )颌下腺。HE染色 ,光镜观察形态学改变。免疫组化染色后 ,计数Bcl- 2及Bax阳性细胞数。结果 随着糖尿病进展 ,颌下腺组织萎缩。Bax阳性细胞数比各同龄对照组明显增多 ,且随病程延长有显著增多趋势。相反Bcl- 2阳性细胞数则明显低于对照组 ,并随病程延长呈下降趋势。结论 抑制细胞凋亡的Bcl- 2表达的降低及促进细胞凋亡的Bax表达的增加提示 ,随着糖尿病发展 ,颌下腺实质细胞有凋亡增强趋势 。 Objective To investigate the morphological changes and expressions of relative cytokine, including Bcl -2 and Bax , in submandibular gland of C57BL /ksj -db/db mutant diabetic mice.Methods The test group included 3,5,6,8,10 month-old db/db mice.The age-matched db/+m mice served as controls. SP immunohistochemistry stains were applied for bcl-2 and bax antibody. Results In db/db diabetic mice, submandibular gland atrophied. Bax and Bcl-2 were expressed in submandibular gland of both groups.Bax immunopositive cells were more significantly increased in diabetes mice than those in control and there was an enhanced trend with diabetic progress.The number Bcl-2 immunopositive cells in submandibular gland were less than the age-match control and showed a descending tendency with the progression of the disease.Conclusion In db/db diabetic mouse, both the down-regulated expression of Bcl-2 and up-regulated of Bax suggest that diabetes induced stronger of apoptosis,which may be the reason of submandibular gland atrophy and dysfunction.
