摘要
用圆二向色性(CD)技术和荧光技术研究伴刀豆蛋白A(ConA)与巨噬细胞膜上受体结合对质膜的影响。结果表明,ConA与膜受体结合后,膜蛋白分子构象、膜分子活动度及细胞膜对Ca^(2+)通透性都发生变化,这对于深入探讨配体与受体作用对膜分子结构的影响和巨噬细胞激活的分子机理均可能有帮助。
The influence of ConA-receptor binding on macrophage membrane structure was studied. Circulardichroism(CD) and fluorescence techniques were used to detect the conformation change of membraneproteins, the mobility of membrane molccules, and the permeability of Ca^(2+) and the concentration ofintracellular free Ca^(2+).The results showed that after ConA-receptor binding, the conformation of macro-phage membrane proteins changed, and the mobility of membrane molecules increased. In addition,the concentration of [Ca^(2+)_i became higher and the permeability of Ca^(2+) enhanced. The above findingsmay be helpful for us to determine the effects of ligand-receptor interaction on membrane molecularstructure and the mechanism of macrophage activation.
