摘要
基于葡萄糖和胭脂红的作用建立了简单、灵敏测定葡萄糖的荧光光谱方法.胭脂红在pH7.0的PBS缓冲溶液中,有一位于586nm的强荧光峰.当葡萄糖加入后,由于与胭脂红结合成荧光性配合物,导致该峰强度显著增强.而且,在2.0×10^(-7)~1.0×10^(-6) mol·L^(-1)浓度范围内,葡萄糖浓度与荧光强度的增加值(ΔI)呈正比关系,检测限是1.22×10^(-7) mol·L^(-1).葡萄糖酸钙片中的淀粉、钙离子等不干扰测定.用本方法测定市售葡萄糖钙片中的葡萄糖,结果与经典邻甲苯胺法测定方法一致.
A simple ,and sensitive fluorescence method was developed for the determination of glucose based on its interaction with carmine .Carmine had one strong fluorescence peak at 586 nm in pH 7.0 phosphate buffer solution (PBS) .As a result of the formation of new fluores‐cence complex between CA and glucose ,the intensity of this peak increased greatly after adding glucose into carmine solution .Moreover ,the difference of fluorescence intensities showed a good linear relationship with the concentrations of glucose in the range from 2.0 × 10-7 to 1.0×10-6mol· L -1with the detechion limit of 1.22 × 10-7 mol · L -1 .The common foreign substances in calcium gluconate tablets such as Ca2+ ,fructose ,galactose and startch did not interfere with the determination .This method is used to determine the concentration of glucose in calcium gluconate tablets with satisfied results ,which is in good agreement with the stand‐ard O‐T oluidine method .
