摘要
本实验研究亚硒酸钠在诱导NB4细胞凋亡过程中对转录因子NFκB活性的影响。用Western印迹法检测细胞核提取物中NFκB亚基P65的含量 ;用形态学观察、DNA琼脂糖凝胶电泳和流式细胞术检测细胞凋亡 ;用MTT法观察对细胞的生长抑制。结果显示 :亚硒酸钠 (≥ 5μmol/L)可抑制NB4细胞生长和诱导细胞凋亡 ,同时亚硒酸钠作用时间和浓度相关性地抑制了转录因子NFκB的激活。结论
In order to evaluate the effect of sodium selenite on the activation of NFκB during selenite induced apoptosis in NB4 cells, Western blot was used to measure the level of P65 in nuclear extraction of NB4 cells treated with sodium selenite to reflect the activation of NFκB; the apoptosis of NB4 cells was determined by morphological observation, DNA ladder electrophoresis and flow cytometry; and MTT test was used to measure the growth inhibition of cells. Results showed that sodium selenite(≥5 μmol/L)suppressed the cell growth, induced apoptosis and inhibited the activation of NFκB in a concentration and time dependency pattern. It was concluded that inhibition of NFκB might be one of the mechanisms in selenite induced apoptosis in NB4 cells.
