摘要
该试验筛选能够利用廉价的乳品废水作为部分替代碳源的高效耐碱型微生物絮凝剂产生菌群。试验采用常规筛选方法,用盐碱地土壤平板划线。从平板上挑取粘性大的菌落接到相应的液体培养基中,摇振荡培养72 h后用高岭土悬浊液测定微生物产生絮凝剂的絮凝活性。最终选出高效微生物絮凝剂产生菌群。将培养基中的商业碳源变换为其他不同类型的商业碳源以及不同浓度的乳品废水,用控制变量法优化其它他产絮条件,最终得出复合菌群的最佳产絮条件:乳品废水的浓度为900 mg/L、最佳乙醇添加比为2%、最佳氮源为蛋白胨、最佳阳离子为Na_2SO_4、最佳初始p H值为11、最佳培养温度为30℃、最佳摇床速度为160 r/min,以及最佳接种量为2%。在最佳产絮条件下微生物絮凝剂的絮凝活性可达94.68%。
The objective of this experiment is to make efficient microbial flocculant with cheap dairy wastewater. Using con-ventional bacterial screening methods and saline soil to line on the plates. Viscous colonies were picked, shaker cultured 72 h.flocculation activity of the microbial flocculant was measured using kaolin suspension and the desire bacteria were selected.The carbon source is converted to other commercial carbons and various concentrations of dairy wastewater. Other floccula-tion conditions were optimized using control variable method. Highly efficient microbial flocculant producing bacteria were fi-nally screened from saline soil. The optimum conditions are as follows: dairy wastewater concentration of 900 mg/L, the added ethanol is 2%, the nitrogen source was peptone,the cation is Na2SO4, the p H value is 11, the culture temperature is 30℃, the shaker speed is 160 r/min and the inoculation amount is 2%. The flocculation activity can reach 94.68% under the above conditions.
