健康小鼠进食绣球菌对腹腔巨噬细胞分泌肿瘤坏死因子α的影响

摘要 测定健康小鼠进食绣球菌对腹腔巨噬细胞分泌肿瘤坏死因子α(TNF-α)的影响.SPF级Balb/C健康小鼠分组进食普通饲料、分别含绣球菌饲料、干品、浸汁饮水或其添加物组合、分别含酵母β-葡聚糖饲料、浸汁饮水或其添加物组合;进食一定时间后,分离腹腔巨噬细胞,使用含质量分数10%胎牛血清的RPMI1640培养液培养,TNF-α试剂盒检测上清液中的TNF-α质量浓度.结果表明,强化进食绣球菌组合(绣球菌饲料+干品+浸汁)的小鼠与对照组或酵母β-葡聚糖组合(酵母β-葡聚糖饲料+浸汁)小鼠相比,体质量不升反降;分离的腹腔巨噬细胞培养6 d,酶联免疫吸附测定技术检测上清液TNF-α,其质量浓度远高于其它两组.研究提示,强化绣球菌进食组合能够提高健康小鼠免疫力,并可能有利于控制体质量. In order to determine the effect of orally administered Sparassis crispa on TNF-α secreting by peritoneal macrophages from healthy mice. SPF grade Balb/C healthy mice were divided into groups for administering mice common feed,feed containing Sparassis crispa,fresh-dried product,product juice or their combination,feed containing yeast β-glucan powder,powder juice or their combination separately. After a period of administering,peritoneal macrophages were isolated and cultured in RPMI-1640 medium containing 10% FBS. Then the concentration of TNF-α in the supernatant was determined using TNF-α ELISA kit. The result show that mice feeded with Sparassis crispa combination( feed containing Sparassis crispa,fresh-dried product and its juice) lost weight,as compared with other two groups,common feed or yeast β-glucan combination( feed containing yeast β-glucan and its juice),which added weight. Furthermore,significantly higher level of TNF-α was determined after 6 d culturing of peritoneal macrophages from mice feeded with Sparassis crispa combination than other two groups. The result suggests that intensified Sparassis crispa combination feeding has capability for improving immunity of healthy mice and in favor of controlling body weight.
