慢性HBV携带小鼠模型评估

Assessment of Chronic HBV Carriage Transgenic Mice

目的评估乙型肝炎病毒(HBV)ayw亚型转基因小鼠是否适用于慢性HBV携带状态的实验研究。方法选取10只该转基因小鼠为模型组,10只同品系正常小鼠为对照组,饲养24周后取材,检测2组HBsAg、HBeAg及HBV DNA的表达,并比较2组ALT、AST浓度,观察小鼠肝脏病理变化。结果模型组小鼠血清HBsAg、HBeAg、HBV DNA均为阳性,ALT、AST水平较对照组轻度升高(P<0.05);模型组小鼠肝组织病理可见局部炎症坏死,对照组小鼠肝脏无明显病理变化。结论实验采用的模型小鼠对HBV免疫耐受,肝组织病理表现与慢性HBV携带者相似,适用于慢性HBV携带状态研究。

Objective: To assess if C57 BL6 J-HBV(ayw) transgenic mice is competent to the research about chronic HBV carriers.Methods: 10 cases HBV transgenic mice were selected as model group, 10 cases corresponding normal mice were selected as control group, both were fed for 24 weeks. ALT and AST of blood serum were determined, hepatic pathology was investigated. Results: Both ALT and AST of Mice in model group were higher than those in control group. About hepatic pathology: there was partly inflammation in model group, and no pathologic changes were observed in control group. Conclusion: C57 BL6 J-HBV(ayw) transgenic mice is immunological tolerance to HBV, its appearance of hepatic pathology is similar to chronic HBV carrier. The mice we used in the test is competent to the research about chronic HBV carriers.
