摘要
采用AFLP技术对来自我国多个省区的 9份假俭草种质资源进行了DNA指纹图谱分析。从 6 4对引物组合中选出 2对引物组合构建了所研究种质的指纹图谱。 2对引物共扩增出 816条谱带 ,多态性比率超过 6 0 8%。遗传多样性分析表明 ,假俭草资源材料间的遗传距离为 0 135~ 0 994。根据以上的AFLP数据进行了聚类分析 ,结果表明假俭草 9个材料分别聚成了四个类型。聚类结果与形态学和同工酶研究结果是一致的 ,表明应用AFLP技术能在DNA分子水平上较好地揭示假俭草的遗传背景和亲缘关系。
DNA fingerprinting analysis of 9 population centipedegrass(Eremochloa ophiuroides,(Munro)Hack.) from several different provinces in China was performed by AFLP technology, and PAGE electrophoresis results were analyzed by using UVIBand Version 99 and SPSS10.0 analysis software. Eight hundred and sixteen fragments were generated by two primer combinations screened from 64 primer combinations and 492 fragments appeared to be polymorphic(60.8%). Genetic distance ranged from 0.135~0.994 between the populations. Based on the AFLP data, cluster analysis was made. Cluster results showed that all the populations studied were clustered into 4 groups. The genealogy from this study is same as that of centipedegrass resulted from the morphological assy and isoenzyme. It is indicated from this experiment that AFLP can successfully reveal the genetic variation and genetic relationships of centipedegrass at DNA molecular.
