摘要
目的 了解人类白细胞抗原 (HLA)系统中最常见、最复杂且最难分型的 B1 5组的多态性情况 ,并探讨其是否影响移植时供者的选择。 方法 对 1 94名随机健康人群用反向 PCR-SSO方法做 HLA-AB分型 ,同时 ,对部分样品作HLA-AB血清学分型 ,以期得到与等位基因对应的血清学特异性。 结果 本研究共得到 7种属于 B1 5组的等位基因 ,包含了 4种血清学特异性。在中国人群 HLA-B1 5组中 ,B* 1 5 0 1占绝大多数 ,B* 1 5 0 2 ,B* 1 5 1 1 ,B* 1 5 1 8,B* 1 5 2 7的分布频率相似。 结论 B* 1 5 2 7较常见 ,其与 B* 1 5 0 1 (两者均为 B62 )在供受者间错配、是否能导致移植物抗宿主反应 (GVHD)等方面 ,需要作进一步的研究。结果提示在移植时对于 HLA-B1 5组如仅做低分辨率分型是不够的。
Objective To know the polymorphism of HLA B15 group that is the most common ,complicated and difficult in HLA systems in Chinese,and to explore its potential effect on transplantation donor selection. Methods The HLA AB locus of 194 unrelated healthy samples were typed with medium to high resolution reverse PCR SSO method,and at the mean time,part of them covered all alleles in the study were retyped with serological method to determine the serological specificity. Results Total seven alleles of B15 group had been detected in this study,and they covered 4 kinds of serological specificity.Among B15 group in Chinese,HLA B *1501 is dominant.However the similar distribution of B *1502,B *1511,B *1518,B *1527 with different specificity should deserve our discreet selection for suitable donor of transplantation. Conclusion Whether mismatch between B *1527 which more common than our former thought and B *1501 both as B62 can induce GVHD or not also deserve further study.All these inferred low resolution typing methods for HLA B15 group in transplantation might be insufficient.
