高效液相色谱法测定利福喷丁含量的方法建立

DETERMINATION OF RIFAPENTINE BY HPLC

本文应用高效液相色谱法,采用 Lichrospher RP-8 色谱柱,以甲醇-乙腈-0.0375mol/L磷酸二氢钾溶液(1:1:1)为流动相,254nm 波长处检测,建立了测定利福喷丁含量的方法。方法学验证试验结果表明:在样品浓度20～180μg/ml 范围内,峰面积与浓度呈良好的线性关系,r=0.9998:样品的平均加样回收率为99.9％,(n=5):精密度、重现性均可满足定量分析的要求;与现行的卫生部部标准(试行)使用的TLC—UV 法比较,结果基本吻合,但更简便、快速。

A HPLC method was established for assay of Rifapentine, using a column packed withLichrospher RP--18 and a mixture of methanol--actonitrile--0. 0375mol/L potassium dihydrogen phosphatesolution (1: 1: 1)as the mobile phase. The detection wavelengh is 254 nm. The result proves that linearrangers are 20～180μg/ml and r = 0. 9998, the recovery is 99. 9 % (n = 5). The method should meet with re-quirements of the assay for Rifapentine and be simple and rapid comparing with the method of TLC.
