摘要
目的:观察MyoD转染后对人横纹肌肉瘤RD细胞分化及生肌调节因子MRF4表达的影响。方法:利用脂质体法将MyoD转染培养中人横纹肌肉瘤RD细胞,原位杂交检测MyoD、MRF4mRNA的表达,观察细胞形态、生长速度变化,免疫细胞化学检测横纹肌肌球蛋白重链及肌动蛋白α-actin表达。结果:转染后RD细胞形态向高分化方向变化,生长受抑制;MyoD、MRF4表达明显增加;细胞肌球蛋白重链和肌动蛋白α-actin表达显著增强。结论:MyoD具有促进RD细胞分化的作用,并可激活另一生肌调节因子MRF4的表达。
Objective:To study the effects of MyoD transfection on the differentiation and expression of Myogenic regulatory factors of human rhabdomyosarcoma RD cells.Methods:The plasmid -MyoD cDNA was transfected into cultured rhabdomyosarcoma cells RD with lipofectin methods.The myogenic regulatory factors MyoD and MRF4mRNA were measured with in situ hybridization and the expressions of myosin heavy (MHC)andα-actin in the cells were assayed with immunocytochemical method.The cell growth and morphologic changes were observed at the same time.Results:The morphology of RD cells tended to highly differentiated and the growth was suppressed in RD cells transfected with MyoD.The expression of MHC andα-actin were significantly increased in RD cells transfected with MyoD,while the expressions of MyoD and MRF4mRNA were up-regulated.Conclusion:Transfection of MyoD can induce the differentia tion of RD cells and up-regulate the expression of MRF4.
