摘要
目的 观察缺氧不同时间新生大鼠心肌细胞凋亡率及心肌细胞 Fas抗原表达的变化 ,以明确 Fas系统是否介导了缺氧诱导的心肌细胞凋亡 .方法 取体外培养的新生大鼠心肌细胞于 95 0 m L· L- 1 N2 ,5 0 m L· L- 1 CO2 孵箱中培养16 h,32 h和 4 8h,造成缺氧损伤的细胞模型 ,分别检测心肌细胞凋亡及 Fas抗原表达的情况 .结果 在缺氧条件下培养16 h,32 h和 4 8h后 ,流式细胞仪检测到心肌细胞的凋亡率分别为 :(2 .9± 0 .5 ) % ,(6 .2± 0 .8) %和 (2 6 .6± 3.0 ) % ;Fas抗原表达率分别为 :(13.6± 5 .1) % ,(2 5 .9± 4 .7) %和 (5 1.0±4 .9) % .结论 心肌细胞凋亡是缺氧时心肌细胞死亡的重要形式 ;心肌细胞凋亡率及心肌细胞 Fas抗原的表达率随着缺氧时间的延长而增高 ;Fas系统参与介导缺氧诱导的心肌细胞凋亡 .
AIM To study the neonatal rat cardiomyocyte apoptosis induced by hypoxia and changes of Fas antigen expression in cardiomyocytes and to verify whether Fas antigen is involved in hypoxia induced apoptosis of cardiomyocytes. METHODS Cultured neonatal rat cardiomyocytes were cultured in an incubator of 950 mL·L -1 N 2 and 50 mL·L -1 CO 2 for 16 h, 32 h and 48 h. Apoptosis of cardiomyocytes was detected by electronic microscope and flow cytometer. Expression of Fas antigen was detected after hypoxia of 16 h, 32 h and 48 h by immunohistochemistry. RESULTS 16 h, 32 h and 48 h after hypoxia, the apoptosis rates of cardiomyocytes were (2.9±0.5)%, (6.2±0.8)% and (26.6± 3.0)% respectively and the expression rates of Fas antigen in cardiomyocytes were (13.6±4.1)%,(25.9±4.7)%和 (51.0% ±10.9)% respectively. CONCLUSION The apoptosis rates of cardiomyocytes and the expression of Fas antigen on cardiomyocytes increase with the time of hypoxia. Fas antigen is involved in hypoxia induced apoptosis of cardiomyocytes.
