大鼠胃黏膜癌变过程中细胞增殖与端粒酶活性关系

Relation of Telomerase Activity and Cell Proliferating in the Process of Carcinomatous Change of Gastric Mucosa in Wistar Rats

目的 探讨大鼠胃黏膜癌变发生发展过程中DNA含量、增殖细胞核抗原 (proliferatingcellnuclearantigen ,PCNA)及端粒酶活性两者之间的关系。方法 采用甲基硝基亚硝基胍 (MNNG)、10 %NaCl混合液及 40 %乙醇灌胃建立大鼠胃癌模型 ,设立正常组、模型组 ,分别采用免疫组化、流式细胞仪及端粒重复扩增微孔板杂交法检测大鼠胃黏膜PCNA、DNA含量及端粒酶活性。结果 本造模方法可诱发出浅表性胃炎 (chronicsuperficialgastritis,CSG)、萎缩性胃炎 (chronicatrophicgastritis,CAG)、肠化生 (intestinalmetaplasia,IM)、异型增生 (dysplasia,Dys)、胃腺癌(gastricadenocarcinoma,GC)等病变。在大鼠癌变过程中 ,随着胃黏膜癌变的发展 ,DNA含量、增殖细胞核抗原表达及端粒酶活性均呈递增趋势 ,并且两者与端粒酶活性具有高度的相关性 (P <0 .0 1,P <0 .0 1)。结论 大鼠胃黏膜癌变发生发展过程中 ,端粒酶的激活与胃黏膜组织细胞的恶性增殖具有重要的相关性。

Purpose: To study the relaitions of telomerase activiy with DNA content, proliferating cell nuclear antigen (PCNA) expression and telomerase activity in the process of carcinomatous of gastric mucosa in Wistar rats. Methods: We studied DNA content, expression of proliferating cell nuclear antigen (PCNA) and telomerase activity using flow cytometer, telomeric repeat amplification (TRAP) - microtiter plate hybridization assay in rats in three groups: normal control, model control and treatment groups with elemene in gastric cancer rats induced by N-methyl-N′-nitro-N-nitrosoguanidine (MNNG), 40 % ethanol and 10 % sodium chloride. Results: The models of various phase of carcinomatous change of gastic mucosa in Wistar rats were estabalished, chronic superficial gastritis (CSG), chronic atrophic gastritis (CAG), intestinal metaplasia(IM), dysplasia (Dys), and gastric adenocarcinoma (GC). With the development of carcinomatous change of gastric mucosa in Wistar rat, the expression of PCNA, DNA content and telomerase activity were increased by degrees, and, each has an important correlation with telomerase activity (P < 0.01, P < 0.01). Conclusions: Telomerase activity has an important relation with expression of PCNA or DNA content in the process of carcinomatous change of gastric mucosa in Wistar rats.
