摘要
目的 :建立一种简便可行的测定环孢菌素 A(Cs A)的高效液相色谱法。方法 :全血经叔丁基甲醚提取后 ,吹干 ,乙氰溶解 ,正己烷进一步脱脂后进样。流动相为乙氰 :水 (80 :2 5 V/V)。色谱法为 YMC-Pack ODS-A 6.0 mm× 1 5 0 mm ,检测波长 2 1 0 nm,柱温 75℃。结果 :均在预测范围内 ,最低检测限为 1 0 ng,最低检测浓度为 2 0 ng/m L,至少在 5 0 -1 5 0 0 ng/m L范围内呈线性 ,平均绝对回收率为 73 % ,RSD均在 1 0 %以内。提取步骤少 ,方法简便 ,快捷。结论 :本法具有灵敏、特异、普通实验室易于开展的优点。用于肾移植术后 Cs A的血药浓度监测 ,效果良好。
Objective: To establish a practical high performance liquid chromatography method measuring the concentration of cyclosporine A (CsA) in patients after renal transplantation. Methods: The drug was extracted from whole blood and analysed using a C 18 (6.0 mm×150 mm) column. Mobile phase was acetonitrile-water(80:25). The flow rate was 1.2 mL /min and the detector was UV-210 nm. The column temperature was maintained at 75℃. Results: Linear range was 50-1500 ng/mL. The detection limit was 50 ng/mL. Analytical recoveries were 73%. RSD values of within-day and between-day were all lower than 10%. Conclusion: The method is simple and accurate. It can have good effects when used in monitoring blood-drug concentration of CsA after renal transplantation.\;
