严重创伤早期大鼠下丘脑HSP70与iNOS表达与分布的时效关系

摘要 目的 研究严重创伤早期大鼠下丘脑热休克蛋白 (HSP) 70与诱导型一氧化氮合酶 (iNOS)转录表达变化分布和意义。方法 采用BIM Ⅲ型生物撞击机致大鼠胸部严重撞击伤 ,并造成单侧股骨骨折 ,运用S ABC免疫组化、原位杂交技术测定下丘脑HSP70与iNOS及其mRNA的转录表达水平。结果 正常对照组HSP70低水平表达 ,在伤后 1小时即开始明显增高 (P <0 .0 5 ) ,6小时达到峰值为正常的 18倍 ,12小时后开始下降 ,2 4小时时主要集中在室旁核内 ;iNOS无基础表达 ,在伤后 1小时开始出现 ,随HSP70同步增高 ,8小时达到高峰 ,较 1小时时增加 13倍。两者相关系数r =0 .976 0 1。结论 严重创伤后早期下丘脑内HSP70和iNOS过度表达 ,在严重创伤应激时下丘脑的损伤与抗损伤机制中起重要作用。 Objective To study the distinct distribution and the time course changes of HSP70 and inducible NOS expressions in the hypothalamua of rats following severe trauma.?Methods Severe traumatic model of severe thorax impact injury with fracture of femur was established in rat. The expressions of HSP70 and iNOS were studied with S ABC histochemistry, and their mRNAs were detected with in situ hybridization.?Results The expression of HSP70 in control group was low basic level. In trauma group,it increased quickly from 1h (P<0.05)after injury and came to the peak at 6 hours(increased 18 fold), the positive cells mainly distributed in the paraventricular nucleus of hypothalamus (PVN) at 24 hours. The positive cells of iNOS could not be discovered until it was one hour after injury,then the number increased alone with HSP70 and reached the peak at 8 hours (increased 13 fold). The coefficient of product moment correlation was 0.97601.?Conclusion Severe trauma stress can cause the overexpression of HSP70 and iNOS in hypothalamus,which plays an important role in the balance of injury and anti injury in the hypothalamus stress reaction.
